如何体外检测高迁移率族蛋白1
高迁移率族蛋白1（High Mobility Group Protein 1），也被称为二性霉素，由HMGB1基因编码，由于在聚丙烯酰胺凝胶电泳中有很高的迁移率而被命名为高迁移率族蛋白1。
下面我们主要以人HMGB1为参考，对高迁移率族蛋白1进行简要的介绍。
蛋白：
HMGB1跟组蛋白一样，HMGB1是最重要的染色质蛋白，在维持核小体稳定及DNA重组、复制、修复及基因转录扮演重要角色。此外，它在感染、炎症及免疫反应中也发挥重要的作用。HMGB1也是炎症损伤的研究热点之一。已有学者提出将HMGB1作为DNA疫苗的佐剂和癌症治疗的靶标。
根据Uniprot上人HMGB1 链接，http://www.uniprot.org/uniprot/P09429，我们制备了三种HMGB1片段，序列选取分别是Met1~Glu215(RPA399Hu01)，Pro9~Arg163（RPA399Hu02），Met1~Glu215（RPA399Hu61）。RPA399Hu01和RPA399Hu01是E.coli中表达的，而RPA399Hu61用的是293F进行表达。这样研究者可根据自己实验目的，选用不同的片段以及表达系统。
抗体：
获得制备的蛋白后，免疫小鼠和兔，获得单克隆抗体和多克隆抗体后经过亲和层析纯化得到高纯度的抗体。单克隆抗体MAA399Hu22，该抗体效价为81w 。
试剂盒:
获得了单克隆抗体和多克隆抗体后，我们研发了HMG1的Elisa试剂盒。试剂盒中包被的是小鼠单克隆抗体，检测抗体是兔多克隆抗体。HMG1在正常和不同疾病模型中的浓度会有很大的变化，武汉云克隆公司也检测了许多样本，根据内部实验结果，我们对46例表观健康的人进行了检测，HMG1的浓度范围为72~582ng/ml，仅供参考。
另外，仅仅是检测试剂盒SEA399Hu，我们目前拥有的文献就有24篇，研究者可以放心使用。
